Can you skip Ms. Marvel?
No. It carries part of the main MCU throughline, and what comes after it assumes you have seen it. The next essential title is Doctor Strange in the Multiverse of Madness. In chronological order, reaching it takes about 46h 38m of essential viewing across 26 titles. See the full MCU order.
Essential as the introduction of a character who leads The Marvels, and it is the warmest thing in Phase Four. It needs nothing before it.
The series is a Jersey City teen comedy first and a superhero show second. Kamala is sixteen, writes fan fiction about the Avengers, is failing her driving test, and wants to go to a convention her parents have forbidden, and the first two episodes are almost entirely about a family. The visual style is the most distinctive Marvel has used on television, with her imagination animated over the real world in doodles and text bubbles.
The strongest material is the fourth and fifth episodes, which travel to Karachi and then into a flashback at the 1947 partition, following her great-grandmother on a station platform. A Disney superhero series stopping to dramatise partition is not something anybody expected, and it is handled with real care.
The weaknesses are the plot's. The antagonists are a faction with a vague objective, a government agency pursues her with no motivation worth the name, and the powers are changed from the comics in a way that annoyed readers. Iman Vellani is the single best piece of casting in Phase Four.
